Hold the item and type /glow (or /i glow), that will give it the Lure enchantment (level 0 i think) and yeah, it will glow.

it isn't possible without an enchantment, the lure 0 enchantment is useless and won't affect the tool

that would only be possible with an item entity, so the thing that appears when you drop it

entity action -> appearance -> set glowing

you can just use that in the drop item event, or right after you spawned an item
